A single injection can reduce the likelihood of suffering a heart attack by 90 per cent and could be used as a vaccination in future for people at risk, research suggetss.
Scientists from the Harvard Stem Cell Institute hope the injection, which only needs to be used once, may be available for patients within 10 years and could be an alternative to statins which are commonly used to reduce the likelihood of heart attacks.
It works by stopping the actions of a gene found in the liver, which prevents the removal of cholesterol in the blood, therefore raising the risk of someone having a heart attack.
The injection has been tested on mice where it was found to reduce concentrations of cholesterol by up to 40 per cent within days of being used on the animals.
Dr Kiran Musunuru, from the Harvard Stem Cell Institute, said: "Heart attack is the leading natural killer worldwide, with one in two men and one in three women past the age of 40 having heart attacks in their lifetimes. Related Articles

"If you had a therapy that targeted the liver, changed the genome - and if it were totally safe - then at least in theory you could think of this therapy as something like a vaccination.”
He said the single injection could be a “permanent alteration” for patients and could reduce the occurrence of heart attacks by between 30 and 90 per cent in humans.
The injection works by targeting PCSK9, a gene which appears primarily in the liver and regulates levels of harmful cholesterol.
It was developed after a team of French scientists discovered in 2003 that people who inherited very high cholesterol levels and corresponding high risk of heart attack had a variant of the gene that was very active.
Another group discovered that around 3 per cent of people had mutations of this gene which has an opposite effect, with lower than average cholesterol levels which reduces their risk of heart attack by up to 88 per cent.
The scientists then attempted to replicate this mutation with a single injection, using gene technology, that would have a permanent impact.
"Our reasoning was that nature has already done the experiment; you have people who have won the genetic lottery," said Dr Musunuru.
"They are protected from heart attack, and there are no known adverse consequences. So that led us to reason that if we could find a way to replicate this, we could significantly protect people from heart attack."
Scientists used a gene editing tool which allows them to erase or disrupt specific DNA sequences. They used this to target PCSK9 so the gene would be disrupted and its activity changed. This gene usually produces a protein which is active in the bloodstream and prevents the removal of cholesterol.
Dr Musunuru added: "Within three to four days of delivering the system into the liver, the majority of the PCSK9 gene copies in all of the liver cells were disrupted, knocked out. And what we hoped to see was much less of the protein product in the bloodstream, which is what we saw.
"The other consequence that we saw was a 35 per cent to 40 per cent reduction in cholesterol levels in these mice, which would translate in humans to a heart attack risk-reduction of as high as 90per cent."
He said once they managed to get the tool into the liver and modify the gene it had a “dramatic effect.”
Dr Musunuru said the single injection is better than using antibodies – a method trialled by other scientists -as these do not last forever and need to be injected every few weeks.
He added: “The main option for reducing cholesterol is statin drugs, such as Lipitor, but many people taking statin drugs every day still have heart attacks. So there is still a great need for other approaches."
The findings are reported in the American Heart Association journal Circulation Research.
